چکیده

In recent years, microfluidics has demonstrated novel methods of culturing and studying cells. Within welldefined volumes comparable to the sample of interest, media, nutrients or toxins may be precisely targeted to individual cells via diffusion or advection, enabled by integrated valves and pumps. Tools, which both enable a repeatable and systematic means of single embryo culture, as well as provide information on metabolic activity of individual embryos, would be of significant utility to the embryologist. To address the need of controlled culture environments and improve embryo health and subsequent positive implantation rates, we have developed programmable microfluidic devices for quantifying metabolite levels in the culture media. Our microfluidic platform provides a high throughput, accurate and repeatable means to non-invasively assess embryo viability, which should be of significant utility to the clinical embryologist.

Behavior of a Metabolic Cycling Population at the Single Cell Level as Visualized by Fluorescent Gene Expression Reporters

BACKGROUND During continuous growth in specific chemostat cultures, budding yeast undergo robust oscillations in oxygen consumption that are accompanied by highly periodic changes in transcript abundance of a majority of genes, in a phenomenon called the Yeast Metabolic Cycle (YMC). This study uses fluorescent reporters of genes specific to different YMC phases in order to visualize this phenom...

A Chick Embryo in-Vitro Model of Knee Morphogenesis

Background: In this feasibility study, a mechanically loaded in-vitro tissue culture model of joint morphogenesis using the isolated lower extremity of the 8 day old chick embryo was developed to assess the effects of mechanical loading on joint morphogenesis. Methods: The developed in-vitro system allows controlled flexion and extension of the chick embryonic knee with a range of motion of ...

Legal Aspects of Iatrogenic Multiple Pregnancy

During Assisted Reproductive Technology (ART) procedures, the transfer of a single embryo dramatically reduces the rate of multiple pregnancy. Proper information, therefore, should be delivered to patients before embryo transfer. In many countries, regulations limit, according to age of the patient, the number of embryos to be transferred. Selective fetal reduction should not be considered as a...

Metabolism of Tissue Cultures

By using radioactive isotopes in tissue cultures, the rate of permeation of substances into cells can be measured independently of concurrent metabolic reactions of these substances. Techniques of obtaining and analyzing data are described. Examples are given using radioactive potassium and phosphorus. Using cultures of chick embryo muscle, turnover time for cell potassium is 6 hours, and for c...

Microfluidic Protocol for Pre-implantation Culture of Single Mammalian Embryos: towards an Optimal Culture Protocol

Microfluidics holds great potential for the field of assisted reproduction techniques (ART), to provide integrated platforms for combined embryo culture and characterization. The development of mouse embryos is not impaired in a microfluidic format: it proceeds faster during the pre-implantation period (< day 4), and gives similar birth rates. More importantly, single embryo culture is enhanced...
